Learn about val­id­a­tion of array-based anti­body-pro­tein inter­ac­tions using Luminex bead arrays

Have you ever struggled with con­firm­a­tion of your pre­vi­ously iden­ti­fied can­did­ate bio­mark­ers? Anti­body val­id­a­tion after ini­tial screen­ing can be tedi­ous and time-con­sum­ing. Togeth­er with our cooper­a­tion part­ner, the Aus­tri­an Insti­tute of Tech­no­logy (AIT) we have con­duc­ted a val­id­a­tion exper­i­ment and tested sera from rheum­at­oid arth­rit­is (RA) patients for inter­ac­tion on engine pro­tein arrays. The res­ults of this exper­i­ment were val­id­ated by using the com­plete cer­ti­fied immu­n­om­ics work­flow imple­men­ted at AIT for screen­ing, con­firm­a­tion and val­id­a­tion of autoantibod­ies.

Learn about the sens­it­iv­ity of engine pro­tein arrays com­pared to com­mer­cially avail­able ELISA using the example of TRIM21

ELISA are com­monly used for dia­gnost­ic pur­poses. There­fore, the ques­tion arises wheth­er engine pro­tein arrays have com­par­able sens­it­iv­ity and spe­cificity to ELISA and wheth­er the inter­ac­tions found with engine pro­tein arrays can be repro­duced in an ELISA screen and vice versa. There­fore, engine per­formed a series of exper­i­ments to invest­ig­ate how their hEXse­lect and Uni­PEx arrays com­pare to a com­mer­cial ELISA for the anti­body effect­or pro­tein TRIM21 (Tri­part­ite motif-con­tain­ing pro­tein 21).

Learn about posttrans­la­tion­al pro­tein modi­fic­a­tions on engine pro­tein arrays, using the example of pro­tein phos­phoryla­tion with pro­tein kinase A

Posttrans­la­tion­al modi­fic­a­tions (PTMs) are enzymat­ic modi­fic­a­tions of pro­teins fol­low­ing pro­tein bio­syn­thes­is in the cell. Many dif­fer­ent PTMs are known, and some can be intro­duced to pro­teins in vitro.
In this study we focus on cAMP-depend­ent pro­tein kinase A (PKA) aim­ing to identi­fy tar­gets and thereby get­ting some more insight into the tar­get pref­er­ence of the kinase.
